I do not believe Conan was ever a stand-up. I believe he was a writer before he got his show. SNL, the Simpsons, etc.

John Stewart is definitely up there. Dane Cook is a hoot. Henry Rollins is a god among men. Stephen Lynch is a genius. Chris Rock is one witty bastard. I'm sure I will think of more.

Robin Williams... hands down. His Improvisational skills are unmatched.

I saw the Inside The Actor's Studio with him, and nearly wet myself laughing. He is a bit of a spaz, and sometimes he disappoints me in films, but in person (like in interviews, or ad-libbing) he is one of the funniest humans I've ever seen.

Jon Stewart's book is bad ass, but he's not much of a comedian as opposed to someone like Seinfeld or Mitch Hedberg.

For me, it's a tie between Jon Stewart and Chris Rock. Because on one hand, you have the funniest smartman, and on the other, you have the smartest funnyman.

Well, Eddie Izzard is obviously the top choice...
But, you can't have Izzard without mentioning all of the Python clan, since he obviously so dearly loves them, and their style.
Have to say Douglas Adams, Simon Jones, etc... the original crew of the HitchHiker radio show...
Agree with Bottle, Rollins is greatly underrated.
Rowan Atkinson deserves a mention.
